UNC Lineberger Seminar Series: High-Resolution Mapping of Cell State and Lineage Dynamics in Cancer

Kathryn Yost, PhD, is a postdoctoral fellow at the Whitehead Institute. Her current research investigates how gene regulation and cell lineage shape cancer progression and antitumor immunity.
As a graduate student, Dr. Yost combined single-cell genomics and computational analyses to uncover the clonal dynamics of T cell responses to immune checkpoint blockade and the differentiation paths of T cell exhaustion. This work also revealed fundamental cancer cell–intrinsic mechanisms, including extrachromosomal DNA clustering in nuclear hubs and widespread enhancer rewiring across human cancers.
During postdoctoral training, Dr. Yost developed PE-tracer, a prime editing– based lineage recording platform, enabling simultaneous tracking of lineage, phenotype, and spatial organization in vivo.
In this seminar, Dr. Yost will share how these innovative tools are illuminating the differentiation, migration, and regulatory circuits of tumor-infiltrating immune cells to uncover new principles governing antitumor immune responses.
